PDGF BB, porcine
Description
Porcine Platelet-Derived Growth Factor-BB (PDGF-BB) is a disulfide-linked homodimer of two B-chains (108 a.a. each). It is stored in platelet α-granules and is released following activation of platelets. PDGF-BB has potent mitogenic effects on various cell types such as cells of connective tissue, bone and cartilage and smooth muscle. It promotes bone formation and soft tissue repair, stimulating wound healing processes. PDGF-BB is involved in the development of the nervous system and may act as a neuronal regulatory agent via nerve regeneration and proliferation. It also plays a role in the epithelial cell development of the respiratory system, cell migration and angiogenesis.

Recombinant MESOkine® PDGF-BB, porcine is produced in the endosperm tissue of barley seed (Hordeum vulgare).

Purity
MESOkine® recombinant PDGF-BB is purified from barley seeds resulting in an extract containing purified recombinant PDGF-BB protein along with co-purified barley seed proteins. The amount of the recombinant PDGF-BB protein is typically in the range of 5-10% of total purified protein content. The exact content of the growth factor is stated on the vials. The co-purified barley seed proteins may serve as stabilizing proteins that can prolong the lifetime of the recombinant protein during proper storage and may enhance the bioactivity of growth factors in standardized bioassays.
Reconstitution & Stability
It is recommended to reconstitute the lyophilized protein in sterile buffered saline or a solution of your choice, preferably to a concentration of no more than 10-100 μg/ml. If needed the reconstituted material may be centrifuged at 3500G for 5 min or filtrated using 0.45 μm filter. The lyophilized protein, though stable at room temperature for few weeks, is best stored at -20°C. Reconstituted protein should be used immediately or stored in working aliquots at -20°C. Avoid repeated freeze-thaw cycles.
Biological Activity
Each batch of MESOkine® growth factor is tested for bioactivity and verified to have comparable activity to a corresponding commercial source of highly purified (>95%) growth factor. The bioactivity of MESOkine® recombinant porcine PDGF-BB was determined by its dose dependent effect on proliferation of 3T3 cells. The ED50 for this effect using PDGF-BB is typically < 5 ng/ml corresponding to specific activity of > 0,2 x 10e6 U/mg. Optimal concentration should be determined for specific applications and cell lines.
Molecular Weight
Recombinant human MESOkine® PDGF-B monomer contains 108 amino acids and a 6 a.a. histidine tag for a total length of 114 a.a. and has a predicted molecular mass of 14.6 kDa. The PDGF forms a disulfide-linked homodimer, PDGF-BB, consisting of two β chains. The recombinant protein migrates with an apparent molecular mass of 17 kDa, with two additional fragments migrating at 8 and 15 kDa in SDS-PAGE.
